PGRN在肿瘤侵袭转移中的研究进展

生长因子PGRN是一种在上皮细胞、免疫细胞、神经细胞和软骨细胞中表达丰富,在人类肿瘤组织中过度表达,并能够促进肿瘤发生发展的分泌性生长因子。人GRN基因突变会引起其编码的分泌性糖蛋白PGRN表达呈半倍剂量性减少,并最终导致中枢神经系统疾病额颞叶退行性变性的发生。PGRN尚在肿瘤、损伤修复、及炎症组织中发挥调节细胞增殖、生存、迁移等功能,随着研究的深入,PGRN在肿瘤侵袭转移中的作用更引起了研究者的关注。研究显示,肿瘤的侵袭转移在肿瘤发生早期即已发生,而生长因子PGRN能够上调肿瘤发生发展中VEGF、MMP-9、MMP-14等因子的表达,促进肿瘤的侵袭转移。本文即对近年来PGRN相关功能及与肿瘤侵袭转移相关性的最新研究进展作出了总结分析。

PGRN (also known as granulin/epithelin precursor(GEP), proepithelin, PC cell-derived growth factor, or acrogranin,is a secretory growth factor abundantly expressed in epithelial cells, immune cells, neurons, and chondrocytes. Mutation of human GRN, the gene encoding the secreted glycoprotein progranulin, results in a form of frontotemporal lobar degeneration, while its overexpression in tumors contributes to tumorigenesis and developing. It is reported that invasion and metastasis of tumor occurs early when tumor develops. PGRN upregulates the expression of VEGF、MMP-9、MMP-14 etc. in tumorigenesis and developing, which accelerates the invasion and metastasis of tumor. Here we summarize its roles in tumor invasion and metastasis.
