目的 研究白藜芦醇对间充质干细胞(MSCs)受照后产生的炎症因子IL-1β的影响,并探讨其机制。方法 将MSCs分为空白对照组、单纯照射组、shRNA干扰组和白藜芦醇组,应用ELISA、Western blot和 RT-PCR等方法检测 IL-1β的分泌和表达。结果 预先给予MSCs 50、100和200μmol/L的白藜芦醇组与未加药组相比,IL-1β胞外分泌显著降低(t=83.34, 24.48, 12.52;P<0.05),同时细胞内IL-1β蛋白表达和mRNA水平也显著降低(t=8.695,14.77,13.9; P<0.05)。照射前沉默Sirt1基因并给予MSCs 200μmol/L白藜芦醇,与照射前单纯给予200μmol/L白藜芦醇相比,辐射引起的IL-1β分泌显著升高(t=18.57;P<0.05),同时细胞内IL-1β蛋白表达和mRNA水平也显著升高(t=10.24; P<0.05)。结论 白藜芦醇可明显抑制辐射诱导的MSCs炎症因子IL-1β的产生,并且Sirt1蛋白在辐射后产生炎症的过程中起到关键的调节作用。

o investigate the effect of resveratrol on IL-1β production in Mesenchymal Stem Cells(MSCs) exposed to radiation, and explore the protective effect and mechanism of resveratrol for MSCs against radiation damage. Mesenchymal Stem Cells(MSCs) exposed to radiation, and explore the role of Sirt1 in radiation protection. Method MSCs were divided into control group, irradiation group and resveratrol group, and pretreatments of different dosage (50μM, 100μM, 200μM) of resveratrol were administrated to MSCs before radiation. The expression of IL-1β was detected using ELISA, western blotting and real time PCR after irradiation. shRNA silence Sirt1 gene transcription in MSCs, and detect the intracellular Sirt1mRNA expression to confirm the gene silencing; And treated with 200μM resveratrol to MSCs before radiation, the expression of IL-1β was detected using ELISA, western blotting and real time PCR after irradiation. Results The level of IL-1β secretion was decreased compared to radiation group (t=83.34, 24.48, 12.52, P<0.05), and meanwhile, the intracellular expression of IL-1β protein and mRNA of resveratrol group were also decreased significantly compared to radiation group (t=8.695,14.77,13.9; P<0.05). The level of IL-1β secretion was increased compared to resveratrol group (t=18.57; P<0.05), and meanwhile, the intracellular expression of IL-1β protein and mRNA of RNAi combine with resveratrol group were also increased significantly compared to resveratrol group (t=10.24; P<0.05). Conclusion Resveratrol could inhibit the production of inflammatory factor IL-1β in MSCs induced by radiation. Sirt1 gene silence can obviously inhibit the radiation protective effect of resveratrol, it suggest that Sirt1 may play a key role in the process of inflammation induced by radiation.
