后肢去负荷对大鼠比目鱼肌AMPK信号通路关键激酶的影响
he Effect of Hindlimb Unloading on AMPK Signaling Pathway of Soleus Muscle in Rats
研究后肢去负荷对大鼠比目鱼肌萎缩程度和AMPK信号通路关键激酶的影响。采用大鼠尾部悬吊法建立后肢去负荷模拟失重动物模型,并将大鼠随机分为2组,即后肢去负荷组和对照组,去负荷处理14d,然后测定大鼠的体重、日增重和饲料消耗量以及比目鱼肌的重量,同时,还测定了AMPK信号通路关键激酶的活性。结果可知,去负荷处理14d后,大鼠比目鱼肌发生了极为严重的肌肉萎缩,模拟失重动物模型建立成功,而AMPK、P-AMPK、ACC、P-ACC的活性明显降低。综上所述,后肢去负荷抑制了AMPK信号通路的关键激酶活性,进而抑制了蛋白合成作用。
In this paper, the effect of unloaded on atrophy extent of soleus muscle of rats and key kinases of AMPK signaling pathway were discussed. Hindlimb unloading model was simulated by tail suspension in SD rats. SD rats were randomly divided into two groups (12 for each), control group and unloaded group. After conducted for 14 days, the body weight, mean daily gain, feed consumption, soleus weight, and key kinases of AMPK were determined. Results shown that severe muscle atrophy of SD rats soleus was formed, after 14 days treatment, the weightlessness animal model established successly, and AMPK, P-AMPK, ACC, P-ACC were decreased significantly. In conclusion, hindlimb unloading decreased protein synthesis by restraining key kinases of AMPK signaling pathway.
