通络救脑注射液对Aβ致脑微血管内皮细胞损伤的保护作用
Protective Effects of Tong Luo Jiu Nao Injection on Brain Microvascular Endothlial Cells injured by β-amyloid
目的 探讨通络救脑注射液及其有效成分对脑微血管内皮细胞的保护作用及机制的研究。方法 建立Aβ致脑微血管内皮细胞(BMECs)损伤模型,用通络救脑注射液及其有效单体成分干预,采用CCK-8染色法测定细胞活性,检测条件培养液中乳酸脱氢酶漏出量,免疫蛋白印迹法检测脑微血管内皮细胞RAGE 的表达。结果 通络救脑注射液可显著提高Aβ损伤的脑微血管内皮细胞活性,减少细胞损伤后LDH的漏出和脑微血管内皮细胞RAGE的表达,与模型组相比, 差异具有统计学意义(P<0.01),通络救脑注射液作用明显优于两个单体成分。结论 通络救脑注射液可提高Aβ损伤的脑微血管内皮细胞的活性,对其具有保护作用,其机制可能与减少血管内皮细胞RAGE 的表达有关,通络救脑注射液的作用优于其单体成分。
Objective To investigate the protective effects and mechanism of Tong Luo Jiu Nao( TLJN)Injection and its active ingredients (including Geniposde and Ginsenoside Rg1), on human brain microvascular endothelial cells (BMECs) injured by β-amyloid (Aβ). Methods Human BMECs were pre-incubated by TLJN and Geniposde and GinsenosideRg1 for 2h, and then injured by Aβ. At different intervals,Cell Counting Kit-8 was employed to assay the activities of BMECs, the leakage of lactate dehydrogenase in the culture of BMECs was examined, and the expression levels of receptor for advanced glycation endproducts ( RAGE ) of BMECs was detected by Western blot method. Results TLJN Injection could significantly improve the activities of human BMECs injured by Aβ, decreased LDH leakage rate,and down-regulate the expression levels of RAGE. The two active ingredients showed different effects at different intervals. The effects of compound TLJN injection were obviously superior to those of two active ingredients. Conclusion TLJN injection has protective effects on the human BMECs injured by Aβ, and its mechanisms is related to the decrease of the expression of RAGE. TLJN injection has better effect than the active ingredients.
