Selection and Identification of a DNA Aptamer Targeted to Cronobacter Sakazakii

ronobacter sakazakii(C.sakazakii ) is a kind of conditional pathogenic bacteria which has a great influence for infants. Four FAM-labeled DNA aptamers demonstrating high binding affinity to Cronobacter sakazakii were identified by a whole-bacterium systemic evolution of ligands by exponential enrichment (SELEX) method and flow cytometric analysis. Aptamer B25P was chosen for further characterization. It displayed a dissociation constant of 38.61±5.624 nM and a high binding affinity .The fluorescence intensity was 81.86% for C. sakazakii and less than 15% for other bacteria, which also characterize its high specificity. Whole-bacterium SELEX is a promising technique for the detection of microbial pathogens that does not require the labor-intensive steps of isolating and purifying complex markers or targets.
