LncRNA ANCR对骨肉瘤细胞生物学特性的影响
Effects of LncRNA ANCR on biological behaviors of osteosarcoma
目的明确ANCR在体外对骨肉瘤细胞的生长、增殖、细胞周期等生物学特性的影响。方法利用shRNA技术敲减骨肉瘤细胞系U2OS,并检测骨肉瘤的生长、增殖、侵袭能力、细胞周期等改变。结果shANCR组的MTT实验OD值较空白对照及阴性对照组显著下降(P<0.01);空白对照组及阴性对照组均有较大克隆形成,但shANCR组克隆形成数较其余2组明显减少,有统计学差异(P<0.01)。Transwell实验发现shANCR组穿过Martigel的细胞数较空白对照组及阴性对照组明显减少(P<0.01)。流式细胞仪检测发现shANCR组的细胞周期停滞于G0/G1期的比例为61.6%,显著高于空白对照组及阴性对照组,S期的比例为21.2%,显著低于空白对照组和阴性对照组,差异有统计学意义(P<0.01)。而G2/M,Sub-G1期则在各组间差别无统计学意义(P>0.05)。结论ANCR敲减后明显抑制了骨肉瘤细胞U2OS生长,增殖和侵袭能力,并且使骨肉瘤细胞阻滞于G0/G1期,显著影响了骨肉瘤细胞的生物学活性。?????
ObjectiveTo investigate the impact of ANCR on biological behaviorofosteosarcoma cell line,including gowth,proliferation, invasion.Methods shRNA technology was used on reduction of osteosarcoma cell line U2OS, The biological behaviors were tested by MTT,Colony formation assay,Transwell and Matrigel assay in the osteosarcoma cells line U2OS with different ANCR expression level to define the function of ANCR. To investigate the cell cycles of U2OS with different ANCR expression level by Flow Cytometer for assessing the influence on osteosarcoma cell cycle. ResultsANCR knockdown was found to decrease the number of U2OS cells compared with the shConcells(P<0.01). Down-regulation of ANCR was found to reduce the number of viable U2OS cell colonies, implyingthat U2OS cellstreated by Lv-shANCR had a colony formation suppression ability compared with the control shRNA cells(P<0.01).The number of U2OS cells crossing the Matrigel coated ?lter in the shANCR group was prominently less than that in the shCon and con group(P<0.01).Inhibition of ANCR expression resulted that osteosarcoma cells were arrested in G0/G1 phase in the shANCR group compared with control group,the rate of G0/G1 was 61.6% (P<0.01).While, the percentages of S phase cells in the shANCR group were markedly decreased,and the rate of s phase was 21.2% compared with control group(P<0.01).Conclusions Decreased ANCR expression inhibited osteosarcoma cell,proliferation and invasion and promoted G0/G1 arrested in vitro,indicating that ANCR mayregulate the biological behavior of U2OS.
