Robo1 在视网膜/脉络膜血管内皮细胞中的作用研究

目的:探讨神经导向因子Robo1在视网膜/脉络膜血管内皮细胞(RF/6A )中的作用。方法:通过siRNA干扰技术降低Robo1基因在RF/6A 细胞中的表达,用RT-PCR 检测mRNA 表达变化,用Western Blot检测蛋白表达变化。采用 MTT,Boyden小室,免疫荧光,流式细胞仪对细胞的增殖,迁移,伸展,细胞周期进行检测。用 Matrigel对RF/6A 血管形成进行检测。结果:siRNA 干扰技术在mRNA 水平以及蛋白水平均有效的沉默Robo1 基因在细胞中的表达。Robo1 siRNA 干扰后RF/6A 细胞的粘附、增殖、迁移均受到抑制,RF/6A 血管形成明显受到抑制。结论: Robo1 siRNA对RF/6A 细胞的细胞增殖、迁移能力的抑制作用以及管道形成能力明显。Robo1基因为临床视网膜新生血管性疾病的治疗提供了新的靶点。
Purpose: To evaluate the roles of Robo1 in the retinal/choroidal vascular endothelial cells (RF/6A). Methods: siRNA technology was used to knock down survivin expression and to study its effects on cells in vitro. Cell proliferation, migration, spreading, cycling, and apoptosis were assessed with the MTT assay, Boyden chamber assay, immunocytochemistry, and flow cytometry. Tube formation by RF/6A on Matrigel was also analyzed.Results:Robo1 siRNA knockdown inhibited cell proliferation, migration. Tube formation by RF/6A cells was also disturbed. Conclusions: Silencing the expression of Robo1 in RF/6A cells inhibited their proliferation, migration and tube formation. This study indicates that Robo1 may be a molecular target for new diagnostic and therapeutic strategies for neovascular retinopathy diseases.
