Synthesis and Photocleavage of DNA by Ru(bpy)32+-(CH2)7-MV2+
本文将电子受体MV2+通过饱和柔性碳链引入到光敏体三联吡啶钌结构中,合成得到新化合物Ru(bpy)32+-(CH2)7-MV2+。利用其分子内高效专一的电子转移特性,研究了它对pBR322质粒DNA的光敏切割活性,通过使用DTT,组氨酸,DMSO、乙醇和甲酸钠等活性氧捕捉剂,充分证明其光敏切割机理为电子转移机理,八元瓜环(Cucurbit[8]uril, CB8)与配合物1的自组装可以有效抑制电子回传,使自组装化合物对DNA的切割能力得到提高。
Ru (II) complex covalently linked with viologen units, Ru(bpy)32+-(CH2)7-MV2+, was synthesized and characterized. Ru(bpy)32+-(CH2)7-MV2+ was found to cleave plasmid DNA strand effectively under irradiation by visible light, due to the photosensitized intramolecular electron transfer from the Ru(bpy)32+ to viologen moieties, the photocleavage of plasmid DNA was observed both in air and under Ar atmosphere. The inclusion between 1 and cucurbit[8]uril can inhibit the backward electron transfer in 1 and then enhance the photocleavage efficiency.
