探讨淫羊藿甙诱导滑膜间充质干细胞(SMSCs)向成骨细胞分化的特性。方法:将第四代SMSCs以5×106接种于5块6孔板,每块板分为空白组、对照组组、淫羊藿甙组,每组2个复孔,2天后换液,加入诱导液。空白组加入含10%胎牛血清(FBS)的 L-DMEM培养基继续培养,对照组在基础培养基中上加入成骨诱导剂(含β甘油磷酸钠10mmol/L、地塞米松0.1umol/L、维生素C50mg/L),淫羊藿甙组为含40ug/L淫羊藿甙的L-DMEM培养基。在第14天取出1块6孔板做碱性磷酸酶染色;第21天取出1块6孔板做茜素红染色,其余3块六孔板分别做碱性磷酸酶、I型胶原和骨钙素免疫组化分析。结果:SMSCs 经成骨诱导液及含淫羊藿甙培养基诱导后细胞从梭形变为类圆形,细胞体积明显增加,14天时细胞集落的表面可见有颗粒状物质分泌,碱性磷酸酶染色为阳性,第13天起可见白色钙结节,21天时茜素红染色阳性、碱性磷酸酶、I型胶原和骨钙素免疫组化见细胞周围染色阳性,表现出成骨细胞的特点。结论:淫羊藿甙能促进SMSCs向成骨细胞分化。?????

Objective: To explore characteristics of Epimedium glycoside inducing synovial mesenchymal stem cells (SMSCs) differentiating into osteoblasts. Methords: The fourth generation of SMSCs were seeded in 5 6-well plates by 5 × 106 ,each plate was divided into 3 groups, blank group, control group and Epimedium glycoside group,each group had 2 holes, two days later the culture medium was changed with inducing fluid. The blank group was added L-DMEM medium containing 10% fetal bovine serum (FBS), the control group was added inducing agent on the basis of the L-DMEM medium (containing β glycerophosphate 10mmol / L, dexamethasone 0.1 umol / L, vitamin C 50mg / L) ,the Epimedium glycoside group was added L-DMEM medium containing 40% Epimedium glycoside. On the 14th day removed a 6-well plate made alkaline phosphatase staining; on the 21st day one of the remaining 6-well plates was made alizarin red staining, the rest three plates was made alkaline phosphatase, type I collagen and osteocalcin immunohistochemical analysis respectively .Results: cellular morphology changed from spindle into oval shape after SMSCs induced by osteogenic inducer and Epimedium glycoside, cell volume increased, on 14th day granular substance was visible on the surface of cell colony, alkaline phosphatase staining was positive, 13 days later white calcium nodules was visible, Alizarin red staining was positive on 21st day ,also alkaline phosphatase ,type I collagen and osteocalcin immunohistochemical staining positive, showing the characteristics of osteoblasts. Conclution: Epimedium glycoside was able to promote differentiation of SMSCs into osteoblast.?????
