近年来,肝纤维化疾病作为慢性肝病向肝硬化发展的必经阶段得到越来越多的重视。肝星状细胞(HSCs)的活化与增值是肝纤维化形成的中心环节。目前针对HSCs的研究多在基因层面,糖谱的研究很少涉及。方法:本实验采取终浓度为2ng/mL的TGF-β1诱导人肝星状细胞系LX-2,半定量RT-PCR检测LX-2细胞系是否发生肝纤维化变化。利用凝集素芯片技术对“静态”LX-2细胞与经TGF-β1诱导发生纤维化的LX-2细胞糖蛋白糖谱进行分析,筛选出糖蛋白糖谱中差异表达蛋白糖链,分析糖蛋白糖链与肝纤维化的关系。结论:TGF-β1诱导人肝星状细胞系LX-2纤维化的方法切实可行;凝集素芯片给出了“静态”LX-2细胞系与“诱导”LX-2细胞系糖蛋白糖谱及其表达差异,分析出一系列肝纤维化疾病相关糖蛋白糖链。

he hepatic fibrogenesis is a kind of disease happens generally, which may develop to hepatic cirrhosis if it couldn’t been diagnosed and treated promptly. The human Hepatic stellate cells (HSCs) activation and proliferation are the center links which the hepatic fibrogenesis forms.At present many research aim at HSCs are mainly in the field of gene,the glycan profile research very little involves. Method: The experiment cultured LX-2 cells in the presence of 2 ng/mL TGF-β1 for 24h.Semi-quantitative RT-PCR examined the LX-2 cell line whether had the hepatic fibrogenesis change.The total proteins of the induced LX-2 cells and its control were extracted, labled with Cy3 fluorescent dye, then incubated with the lectin microarray. Glycoprotein Glycan profiles were analysed to screen differently expressed Glycans, and get the relations of glycoprotein glycan and hepatic fibrogenesis disease. Conclusion: TGF-β1 could be used to induce LX-2 cells; The lectin microarray could give the “quiescent” LX-2 cells and the “activated” LX-2 cells induced by TGF-β1 glycoprotein glycan profiles and differences in glycan-profiles.
