雌激素促大鼠急性心肌梗死后心肌血管生成及受体介导机制*
Estradiol promote myocardial angiogenesis in a rat model of acute myocardial infarction through estrogen receptors.
目的 探讨雌激素对去卵巢大鼠心肌梗死后心脏雌激素受体表达及血管生成的影响。方法 结扎去卵巢Sprague-Dauley(SD)大鼠左冠状动脉制作急性心肌梗死模型,给予不同剂量苯甲酸雌二醇干预治疗。放免法测定心梗前和心梗后四周血清雌二醇水平,免疫组化法测定心梗四周后心肌组织中新生毛细血管的数量,Westernblot测定心肌组织中雌激素受体α、β的表达。 结果 去卵巢后大鼠血清雌二醇水平显著降低,不同剂量雌二醇干预后各组血清雌 二醇水平升高。去卵巢心肌梗死组新生血管数目显著低于保留卵巢急性心肌梗死组及去卵巢后雌激素干预各组,其中,雌激素替代组新生血管数目最高。各实验组雌激素受体α、β蛋白表达均有差异,雌激素替代组表达最高,去卵巢组表达最低。结论 体内雌激素同心梗后血管再生有关,雌激素替代治疗有助于增加去卵巢大鼠心梗模型的心肌血管生成,其机制可能是通过调节心脏雌激素受体α、β介导,且ER-α可能发挥主要作用。
Objective To study the influence of estradiol on myocardial angiogenesis and the expression of estrogen receptors in a rat model of myocardial infarction .Methods Acute myocardial infaction(AMI) model was bulit by ligation of Left anterior descending coronary in ovariectomized Sprague-Dauley rats..They were treated with different doses of estradiol Benzoate..Estradiol concentration in serum was measured by Radioimmunoassay before AMI and 4 weeks later,microvascular density in myocardium was evaluated through detection of expression of Ⅷ factor by Immunohistochemistry and estrogen receptorαandβprotein in the myocardium were deteced with western blot at 28 days after AMI. Result Estradiol levels in serum significant degraded after ovariectomized and heightened in the groups with different doses of estradiol. microvascular density in myocardium 4 weeks after AMI in ovariectomized group were obviously lower than acute myocardial infarction group and ovariectomized estrogen therapy groups, estrogen replacement therapy group was the highest. The expression of estrogen receptorαandβprotein in the myocardium showed significant different. estrogen replacement therapy group was the highest and ovariectomized group was the lowest .Conclusion Estrogen improves myocardial angiogenesis in rats after AMI,this efffect could related to the expression of estrogen receptorαandβin myocardium and ER-αplays a major role in this process.
