目的:通过免疫组化的方法探讨复方浙贝颗粒联合阿霉素对KG-1a细胞移植瘤PI3K/AKT信号通路的影响。方法:将KG-1a细胞接种BALB/c-nu裸小鼠腋前皮下构建移植瘤动物模型,给予复方浙贝颗粒灌胃联合阿霉素腹腔注射,实验结束后处死小鼠,剥离肿瘤,将瘤块组织制成切片,通过免疫组化结合Image Pr0Plus 6.0免疫组化彩色图像分析,观察复方浙贝颗粒联合阿霉素对KG-1a移植瘤细胞PI3K/AKT相关蛋白表达的影响。结果:与阿霉素组比,高剂量CZBG可以明显抑制mTOR蛋白的表达(P<0.05),CZBG各剂量组均可以抑制NF-κB蛋白的表达(P<0.05)。结论:CZBG可以协同阿霉素下调mTOR及NF-κB的表达。

Objective:To investigate the effect of Compound Zhe Bei granule(CZBG)combined with doxorubicin on the expression of mTOR ,PI3K,NF-κB and PTEN in KG-1a cell line transplantation tumors in Mice.Methods:Subcutaneous tumor xenograft model is established by injecting the acute myeloid leukemia cell line KG-1a in the axillary flank of BALB/c-nu nude mice.Drug-combination of CZBG intragastric administration and doxorubicin intraperitoneal injection (i.p.) is given to the BA LB/c-nu nude mice.The tumor xenografts are made into slice after the dissection, and the expression of mTOR ,PI3K,NF-κB and PTEN in KG-1a cell line tumor xenografts are detected by immunohistochemical technique. The optical density(OD) of mTOR ,PI3K,NF-κB and PTEN in KG-1a cell line tumor xenografts are measured by Image PmPlus6.0.Results: Compared with ADM group , high-doseage CZBG can inhibit the mTOR expression obviously(P<0.05) ,mid-doseage and high-doseage CZBG can both inhibit the NF-κB expression(P<0.05) .Conclusion: CZBG can collaborate with doxorubicin to reduce the expression of mTOR and NF-κB.
