基于冷冻电镜三维重构技术的Bm-CPV结构与转录机制研究
Structure and Transcriptional Mechanism Resaerch of the Bm-CPV based on the Cryo-EM 3D Reconstruction
质型多角体病毒(Cytoplasmic polyhedrosis virus,BmCPV),属呼肠孤病毒科(Reoviridae),质多角体病毒属(Cypovirus),是一种典型的双链RNA病毒。病毒无囊膜,具单层衣壳。病毒外形为对称的正20面体,直径约为50~65nm,病毒衣壳上具有12个顶点。CPV一般经口传染,也可通过血体腔感染。感染CPV病毒的宿主发病与死亡周期缓慢,约3~18 d不等,部分CPV宿主的死亡周期则更长。
s the simplest single-layer capsid dsRNA virus of Reoviridae family, cytoplasmic polyhedrosis virus (CPV)-the representative virus-is always the most significant point and has great significant in Reoviridae virus study. By cryo-electron microscopy and three dimension reconstruction, the structure of the CPV have been determined: a icosahedral partical with twelve vertices, and there are ten transcriptional enzyme complexes(TECs) organized with ten dsRNA in a specific. Capsid is consists of sixty asymmetric unit. Asymmetric unit contains one turret protein(TP), two large protrusion protein(LPP) and two capsid shell protein(CSP). Turret is consist with five channels formed with TP, and each TP contains three active site domains. When MT-2 binding a S-adenosyl-L-methionine(SAM), the virus capsid expand and active the transcription process, and the nascent mRNA will across the channel and begain the capping process. This review summarizes some professional papers about CPV, and introduce the mechanism of CPV structure, transcription and capping process,hope can help other researchers in their study of the CPV.
