甘薯长喙壳病菌对三种寄主的致病性与ITS分析

已有研究表明甘薯长喙壳菌具有明显的寄主专化性。本试验为了探明甘薯长喙壳菌对石榴、甘薯和芋头三种寄主的致病性,分别用针刺、浸根接种方法进行了原寄主/交互接种,同时进行rDNA-ITS序列分析。结果表明从三种寄主上分离的病原物都对原寄主有致病性;甘薯分离物对石榴和芋头均不能侵染,表现出明显的寄主专化性.然而,石榴和芋头分离物则能够相互侵染。为此,对石榴、甘薯、芋头的分离物进行ITS扩增,得到600bp左右片段。经测序并在NCBI上进行BLAST比对,发现石榴菌株与芋头菌株的同源性为100%,而与甘薯菌株的同源性为97%。通过DNAMAN对该六个菌株进行同源序列比较,发现石榴与芋头菌株的碱基差异较少,而甘薯与石榴和芋头菌株的碱基差异较大。

It was recorded that Ceratocystis fimbriata was a host-specialization.This experiment aims to study pathogenicity of Ceratocystis fimbriata on pomegranate, sweet potato and taro, with pricking and dipping root method to original/cross host inoculation. And do rDNA-ITS sequence analysis as well. The results showed that the isolates have the pathogenicity to their three own host respectively; The isolates from sweet potato can not infect that pomegranate and taro with host -specialization. However, The isolates from pomegranate and taro can infect each other. The ITS(internal transcription spacer )primer pair ITS1 and ITS4 were used and amplified the fragment around 600bp. Analysis of the ITS sequence data showed that the isolates were 100% homologous between taro and pomegranate isolates, but 97% homologous from sweet potato to both taro and pomegranate isolates by BLAST analysis. The comparison results by DNAMAN showed that the taro and pomegranate isolates almost the same, more difference between sweet potato to taro and pomegranate isolates.
