目的 探讨大鼠Islet-1(Insulin gene enhancer binding protein 1)基因是否有诱导NSCs向胆碱能运动神经元分化的能力。方法 用Islet-1基因重组逆转录病毒载体转导NSCs后,用免疫荧光化学染色检测Islet-1在NSCs内的表达;体内、体外实验观察导入Islet-1的NSCs向乙酰胆碱转移酶(choline acetyltransferase, ChAT)阳性细胞分化的情况。结果 在体外分化实验中发现,转导Islet-1基因的NSCs 向胆碱能运动神经元分化的细胞数明显比对照组为多,体内移植实验发现导入Islet-1的NSCs在体内可以向ChAT阳性细胞分化。结论 Islet-1基因有诱导NSCs向胆碱能运动神经元分化的能力。

Objective To explore whether Islet-1 gene could induced NSCs differentiate into cholinergic neurons. Methods Cultured NSCs infected with medium including Islet-1 recombinant retroviral expression vector. The protein expression of Islet-1 gene in NSCs was detected by immunofluorescence. The ability of NSCs differentiation into ChAT positive cells was observed in vivo and vitro. Results The numbers of ChAT positive cells were significantly increased in group of NSCs modified with Islet-1 compared with control group in vitro. NSCs modified with Islet-1 could differentiate into ChAT positive cells when they grafted into corpus striatum of the adult rat brain. Conclusion Islet-1 gene could induce NSCs differentiate into cholinergic neurons.
