埃博拉病毒VP40蛋白抑制剂的虚拟筛选
Virtual Screening for VP40 Inhibitors of Ebola Virus
埃博拉病毒是一种具有极大公共危害性的流行病毒,感染人后有50%到90%的死亡率。埃博拉疫情严重的一个重要原因是缺乏药物和疫苗,因此相关的药物和疫苗的开发受到特别关注。本研究探用老药新用的方式,应用计算机虚拟筛选方法,以埃博拉VP40蛋白的活性口袋作为作用靶点,分析已获FDA批准的上市药物分子库中是否存在能够与埃博拉VP40蛋白发生相互作用的药物分子。结果筛选出五个化合物,在分子对接中最具有与VP40相结合的潜质,值得进一步通过实验来验证。本研究可为后续抗埃博拉药物研究提供参考。
Ebola virus is known as the pandemic and deadly influenza virus ,which causes Ebola hemorrhagic fever in human and primates. It was becoming endemic in middle Africa with the motality between 50% and 90%. The virus is considered as one of the most severe virus harmful to humans, for there are not effective drugs and vaccines. Therefore, to study the prevention and treatment of ebola virus is of great significance. Our work is based on the thought of old drugs used in new ways by computer virtual screening method to search inhibitors. We aimed to screen small molecules existing in FDA-approved drugs which could bind with ebola VP40 protein. In this study, we selected five small drug molecules which are promising to bind with VP40 by counting binding score and shape complementarity. We expect the study of this paper can provide useful information for the subsequent ebola drugs research.
