醋栗miRNAs的鉴定及表达研究

Identification and expression analysis of microRNAs of Ribes nigrum

MicroRNAs(miRNAs)是一类内源性单链、长约21nt的非编码RNAs,在植物生理及代谢过程中起着至关重要的作用。虽然目前在很多植物物种中鉴定了miRNAs,但醋栗作为一种重要的小浆果经济作物目前尚未见到miRNA方面的报道。本研究利用基于EST的比较基因组学方法对醋栗中的miRNAs进行了鉴定。根据一系列的筛选标准,在8496条醋栗ESTs序列中鉴定出3个miRNAs,分别命名为ram-miR5021、rni-miR5021和rni-miR5185。随后对产生rni-miR5021和 rni-miR5185的MIR基因进行克隆并转入到拟南芥中,结果miR5021和miR5185在转基因拟南芥中表达量明显增加。这表明我们鉴定的这2个醋栗MIRs基因确实在能够在植物体内产生相应的miRNA。进一步利用qRT-PCR方法对这2个醋栗miRNAs的前体及成熟体的组织表达特异性进行了分析。这些发现将大大促进未来对醋栗miRNAs功能及调控机制的研究。

MicroRNAs (miRNAs) are a class of single-stranded, endogenous, small non-coding RNAs, which serve as regulators of gene expression to perform important functions in plant biological and metabolic processes. Although numerous miRNAs have been identified in many plant species, miRNAs still keep unknown in Ribes, which are economically important small fruit crops. In this study, an EST-based comparative genomics approach was employed to identify miRNAs from Ribes. According to a series of filtering criteria, 3 miRNAs were identified from 8,496 Ribes ESTs and were designated as ram-miR5021, rni-miR5021 and rni-miR5185. Subsequently, the 3 MIR genes encoding rni-miR5021and rni-miR5185 were cloned and transferred into Arabidopsis, and the accumulations of miR5021 and miR5185 were significantly produced in transgenic Arabidopsis, suggesting that these putative MIR genes identified from Ribes were able to generate their corresponding miRNA in vivo. Furthermore, the expression patterns of the Pre-miRNAs and accumulations of mature miRNAs in different tissues of Ribes were examined by qRT-PCR approach. These findings will greatly facilitate future studies on the functions and regulatory mechanisms of miRNAs in Ribes.
