摘要
为比较牡丹单花品种‘梨花粉’、有侧花品种‘姊妹游春’、‘云鄂粉’花芽分化进程、形态特征差异及花芽分化期内源激素变化,为牡丹长花期育种提供参考依据。该研究通过解剖和石蜡切片,观察‘梨花粉’、‘姊妹游春’、‘云鄂粉’花芽分化过程,酶联免疫吸附分析法测定不同分化阶段‘梨花粉’及‘姊妹游春’的生长素(IAA)、脱落酸(ABA)、赤霉素(GA3)及玉米素核苷(ZR)含量的变化规律。结果表明:(1)‘梨花粉’花芽分化仅 6 个时期;有侧花品种‘姊妹游春’、‘云鄂粉’花芽分化为 11 个时期,顶花原基先分化,侧花原基后分化。(2)叶原基分化期至苞片原基分化期,‘姊妹游春’和‘梨花粉’花芽中 ABA、GA3 及 ZR 含量均升高,IAA 降低;‘姊妹游春’侧花原基分化起始阶段,ABA、GA3 含量降低,ZR、IAA 升高。(3)‘梨花粉’ABA/IAA 比值在形态分化期时最低。以上结果显示,有侧花品种花芽分化比单花品种早,分化时间长,顶花和侧花部分花器官分化期重叠。较高水平的 ZR、IAA 以及较低的 ABA、GA3 有利于启动侧花原基分化。
Abstract
The flower bud differentiation process, morphological features and endogenoushormone changes during single-flowered Paeonia suffruticosa cultivar Lihua Fen and lateralfloweredcultivars Zimei Youchun and Yune Fen were compared in order to provide areference for the long florescence breeding of tree peony. The flower bud differentiation of LihuaFen, Zimei Youchun and Yune Fen were observed by paraffin sections. The contents of auxin (IAA), abscisic acid (ABA), gibberellin (GA3) and zeatin nucleoside (ZR) in Lihua Fen andZimei Youchun at different differentiation stages were determined by enzyme-linkedimmunosorbent assay (ELISA). The results were as follows: (1)The flower bud differentiation ofLihua Fen only were 6 stages and that of Zimei Youchun and Yune Fen were 11 stages. Theapical flower primordium differentiated earlier than the lateral flower primordium. (2) Thecontents of ABA, GA3 and ZR of Zimei Youchun and Lihua Fen increased during the leafprimordium differentiating to the bract primordium, while the contents of IAA decreased. As forZimei Youchun, ABA and GA3 contents decreased at the initial stage of lateral flowerprimordium, while ZR and IAA increased. (3) The ABA/IAA of Lihua Fen was the lowest atmorphological differentiation stage. The study reveals the lateral-flowered cultivars have earlierand longer flower bud differentiation compared with the single-flowered cultivar, parts of thefloral organs differentiation stage of apical and lateral flowers coincided. Higher ZR and IAAlevels and lower ABA and GA3 levels are beneficial to initiate lateral flower primordiumdifferentiation.关键词
