抗菌肽基因转化大豆的研究
Study on Transformation of Antimicrobial Peptides Gene into Soybean via
用农杆菌介导法将抗菌肽基因转入大豆胚尖外植体,用PCR扩增和绿色荧光蛋白表达情况来检测转基因植株。60 mg/L的卡那霉素可以对转化植株进行有效的筛选;在共培养时添加200 μmol/L乙酰丁香酮有利于抗菌肽基因的转化;在生根培养基中不添加卡那霉素对再生植株进行二次筛选更有利于转化苗的成活。经检测证明抗菌肽基因已转入大豆植株中,并得到了表达。
antimicrobial peptide gene was transformed into soybean via agrobacterium-mediation. PCR amplification and expression of green fluorescent protein was used to detect transgenic soybean plants. The 60 mg/L kanamycin could effectively filter the transformed plants, and 200 μmol/L acetosyringone in coculture medium was benefit for improving the gene transduction efficiency. The kanamycin in rooting medium is harmful to shoots root. The PCR amplification and and expression of green fluorescent protein proved that the antimicrobial peptide genes has been transformed into soybean and expressed.
