Effects of whole soybean powder (Glycine max.L) on lipid metabolism in the mice fed with a high-fat diet

本文对全豆粉改善高脂日粮小鼠血脂代谢的可能机制进行了研究。将6周龄C57BL/6J雄性小鼠随机分为4组:基础对照组(AIN-93 M)、高脂对照组(HF)、高脂+脱渣大豆组(HFS)和高脂+全豆组(HFWS)。喂养8周后处死小鼠,分别测定血清中总胆固醇(TC)、甘油三脂(TG)、高密度脂蛋白胆固醇(HDL-C),显微镜观察肝脏组织切片,RT-PCR测定肝脏组织SREBP-2、CYP71a、LXRα、HMGCR的基因表达。结果显示,与高脂日粮小鼠相比,HFWS组小鼠体重有所增加,血清中TC,TG含量明显降低(P<0.05),小鼠AI指标显著升高(P<0.05)。RT-PCR结果表明,HF组肝脏CYP71a和LXRα表达显著下降,SREBP-2和HMGCR表达显著上升;与HF组小鼠相比,HFWS组和HFS组均能够显著上调LXRα和CYP7a1的mRNA表达(P<0.05),下调SREBP-2、HMGCR的表达(P<0.05)。结果表明全豆粉降血脂效果比脱渣大豆组更加显著,这可能与全豆粉膳食纤维比脱渣大豆粉高有关。

his paper was designed to investigate the beneficial effects of the whole soybean powder, compared with the processed soybean powder(soybean milk without okara in traditional soybean production), on the lipid metabolism in C57BL/6J mice fed with a high-fat(HF) diet. The mice were randomly divided into four groups: control group (AIN-93 M), high-fat group (HF), high-fat + powdered soybean(HFS) and high fat + powered whole soybean (HFWS).The mice were sacrificed after eight weeks and the concentration of total cholesterol (TC), triglyceride (TG) and high density lipoprotein cholesterol (HDL-C) in serum were measured. Liver histological analysis was performed by microscopy. The mRNA expression of SREBP-2, CYP71a, LXRα and HMGCRin liver were measured by RT-PCR. Compared with the high-fat diet group, the body weight, serum TC, TG level of HFWS group were significantly decreased (P<0.05), while atheroclerosis index (AI) was significantly increased (P <0.05). RT-PCR result showed that the expression of CYP71a and LXRα were significantly down-regulated, the expression of SREBP-2 and HMGCR were up-regulated in HF group. Compared with HF group, HFWS and HFS were significantly increased the mRNA expression of LXRα and CYP7a1 (P <0.05), decreased the expression of SREBP-2 and HMGCR (P <0.05). The results implied that the whole soybean have more beneficial effects on the control of serum lipid than the processed soybean. It is possible that whole soybean is rich of dietary fiber than processed soybean.
