mRNA差异显示技术及其研究进展
mRNA Differential Display and it’s current research advances
控制生物形状的基本单位是基因,研究细胞的基因表达一直是分子生物学的重要课题之一。不同细胞间基因表达的差异决定了生命活动的多样性,如发育与分化、内环境稳定、细胞周期调节等。近年来,随着DNA重组技术的发展,已有数万个人类基因被克隆分析(人类约30000~35000个基因),要从如此众多的表达基因中找出引起细胞生理或病理变化的基因并非易事。mRNA差异显示技术(DDRT-PCR)的出现将有可能在一定程度上起到关键作用。本文主要从该技术的原理、优缺点及其改进、应用现状和前景等方面作了阐述。
mRNA differential display was established by Liang and Pardee in 1992 for the purpose of displaying the mRNA differences between two tissues. The technique makes it possible to detect the genes differential displayed between specified cells and tissues at the level of mRNA. All the mRNA changes could be displayed side by side as cDNA band differences by mRNA differential display and the genes corresponding to these differential mRNA could be obtained. Most cell activities with changes at the transcriptional level can be investigated and, thus, it can be applied to addressing such topics as cell differentiation, development, cell cycle and response of cells to their environment. The paper mainly focuses in the principle, advantages, disadvantages, modification, application and prospects of the mRNA differential display.
