Preparation of Monoclone Antibodies to Toxoplasma gondii Surface antigen 1

SAG1蛋白在不同毒力虫株都具有良好的功能保守性,因此制备SAG1的相应识别抗体适用于不同毒力虫株的研究。以原核表达的SAG1重组蛋白为免疫原,免疫BALB/c小鼠, 取脾与SP2/0细胞融合,经四次亚克隆,获得了8株稳定分泌、能与重组SAG1蛋白结合的单克隆抗体分泌株:2D8、2G2、3G12、4B7、4H8、5A2、6E1、7C1。以提取的虫体可融性蛋白和SDS溶解的虫体蛋白ELISA鉴定这8株抗体,得到一株4B7可识别虫体蛋白。经测定,该株抗体亚型为IgG 1,滴度为1:3200,经western blot鉴定4B7与重组SAG1有良好反应性。

SAG1 is a conservative protein, therefore the antibody identified SAG1 can be used as a tool in diferent strains. In this study, BALB/c mice were immunized by recombinant SAG1 protein expressed from prokaryotic system, then fused spleen cells with SP2/0. Eigtht McAbs strains against SAG1 were obtained after subcloned four times, and they were named as follows: 2D8、2G2、3G12、4B7、4H8、5A2、6E1、7C1. Soluble proteins and unsoluble proteins dissoved by SDS from toxoplasma gondii were applied to identify these eight McAbs strains by ELISA, and results showed that 4B7 was able to recognise nature proteins. Furthermore, we determined 4B7 was IgG 1 with a title of 1:3200 and had a decent reactivity with recombinant SAG1 protein by western blot.
