pM蛋白与IgY的互作研究

目的:验证pM蛋白与IgY的结合关系,并探明其亲和力大小。方法:使用Pull down、蛋白印迹法、酶解法以及表面等离子共振的方法探索pM蛋白与IgY的结合机制。结果:pM蛋白与IgY存在较强的结合关系,并且pM蛋白与IgY的结合主要依赖于IgY的Fab区域。结论:pM蛋白能够作为IgY纯化的配体使用。
Pull-down assay showed protein pM could capture chicken IgY, duck IgY and duck IgYΔFc, a unique IgY form lacking of CH3 and CH4 and not existed in chicken. Furthermore, the absorbents of IgY hydrolysate (by papain digestion) showed protein pM can bind with IgY-Fab instead of IgY-Fc. Moreover, Western blot assay showed protein pM could bind with IgY-scFv. The affinity of protein pM and IgY-scFv was 1.922E-10M detected by surface plasmon resonance (SPR). These results confirmed protein pM can bind with chicken IgY, as well as IgY from Anatidae birds, by blocking the variable region of IgY.
