电穿孔法在衣藻叶绿体转化中的应用

莱因哈德衣藻是真核细胞叶绿体生物学研究中的一种重要模式生物,有关的研究一直受到其转化方法的限制。目前用于衣藻转化的方法主要有玻璃珠法、农杆菌介导法、粒子轰击法和电穿孔法。其中电穿孔法是核转化最有效的方法,而叶绿体的转化一直停滞不前,因为没有一种有效的方法既可以穿过细胞壁和三层膜结构将外源DNA发送到叶绿体中,又不至于严重损伤细胞。本研究中,我们用电穿孔的方法将一个结构单元的DNA导入到衣藻的叶绿体中,建立了一套简单高效的叶绿体转化方法。试验数据说明:电穿孔法也是衣藻叶绿体转化的一个高效手段。

hlamydomonas reinhardtii was an important model organism in the world of eukaryotic cell chloroplast biology. Up to now, several protocols can mediate the transformation in C.reinhardtii, include glass beads, Agrobacterium tumefaciens, electroporation and particle bombardment system. Electroporation is the most efficient method for nuclear genome transformation. However, the stable transformation of C.reinhardtii chloroplast was long hampered by non-efficient methods to deliver foreign DNA across the cell wall and three membranes. At the same time, the recipient cells were not caused severely mechanical damage for the manipulation. In this research, we introduced a construct into recipient cell by electroporation and established a simple high-efficiency strategy for the transformation of C. reinhardtii chloroplast. Our data indicated electroporation is a simple method for high-frequency chloroplast transformation of Chlamydomonas.
