多功能纳米载体CDP/CDPF用作基因载体功能的研究
Study on the function of multi-functional nano carrier CDP/CDPF as gene carrier
目的:探讨多功能纳米载体CDP和CDPF的生物相容性及转染能力。方法:选取143B细胞系进行实验,通过琼脂糖凝胶电泳实验检测CDP和CDPF对AEG-1 siRNA的复合能力;采用MTT法检测CDP和CDPF对143B细胞的毒性,通过荧光倒置显微镜和流式细胞仪检测CDP和CDPF运载AEG-1 siRNA的能力。结果: CDP/siAEG-1和CDPF/siAEG-1复合物在N/P值大于8:1时,能完全包裹AEG-1 siRNA;CDP/siSCR和CDPF/siSCR在100 μg/mL浓度下都未出现细胞毒性;CDPF/siAEG-1复合物在N/P值为20:1时表现出最高的转染效率(98.7% ± 3.1%),高于CDP/siAEG-1在N/P值为20:1时的转染效(75.1% ± 2.6%)和Lipofectemine 2000/siAEG-1复合物的转染效率(97.1% ± 3.8%)。结论:CDP和CDPF具有良好的生物相容性和很好的运载 siRNA的能力。
Objective:To explore the biocompatibility and transfection ability of multi-functional nano carriers of CDP and CDPF. Methods:143B cell line was used for research.Using Gel retardation assay to detect CDP and CDPF composite capability of AEG-1 siRNA, MTT assay was used to detect CDP and CDPF toxicity in 143B cells, the ability of AEG-1 of CDP and CDPF to carry siRNA was detected by fluorescence inverted microscope and flow cytometry. Results:Both CDP and CDPF started to form complete complexes with siRNA at an N/P ratio of 8; Even at high concentration of CDP and CDPF complexes (100 μg/mL), showed no obvious negative effect on cell viability; Cells incubated with CDPF/siAEG-1 (N/P:20) complexes exhibited almost similar efficiency (98.7%± 3.1%) of transfection compare to Lipofectamine 2000/AEG-1 siRNA complexes (97.1%± 3.8%). Conclusions: CDP and CDPF could be used as a no obvious cytotoxicity small interference RNA transfer vector and high efficient non-viral vector for gene delivery to143B cells.
