水稻滇粳优1号近等基因系的鉴定
Identification of near-isogenic lines from Dianjingyou 1
本研究利用567个分布于水稻基因组上的SSR分子标记,鉴定滇粳优1号及其12个抽穗期表型变化的近等基因系材料。通过比较多态性位点,寻找到近等基因系中可能含有的外源置换片段,分析置换区域内已知的抽穗期基因,最终对近等基因系中抽穗期变化的主要成因进行分析。经分析鉴定,DJ1和DJ2两个近等基因系材料来源一致,且野生稻来源的隐性dth3基因延迟了近等基因系的抽穗;而Y4284因含有野生稻来源的Hd6隐性等位基因而表现出提前抽穗。Y8266和Y8296含有同样的置换区段,其间包含有HGW和Hd1基因;这些基因是否是影响抽穗期变化的原因还有待于进一步确认。此外,其他置换系遗传背景较杂,需进一步回交。这些材料的鉴定不仅有助于挖掘新的抽穗期基因相关材料,而且为构建同一背景下不同基因的双突或三突,研究不同基因之间的相互作用奠定了基础。
In this study, a total of 567 SSR molecular markers distributed in the rice genome were used to identify Dianjingyou 1(DJY) and twelve near-isogenic lines (NILs), which show divergent heading date from Dianjingyou 1. By comparing the polymorphic loci, searching the introduced fragments in NILs, analyzing the known heading date genes within the fragments, the cause of the change in heading date were analyzed. As the results indicated, DJ1 and DJ2 derived from the same source, and the recessive gene dth3 from wild rice may delay their heading date; whereas Y4284 flowers earier than DJY due to the recessive allele in Hd6 loci. In addition, Y8266 and Y8296 contain the same exogenous sections, HGW and Hd1 were located in; the role of these genes needs to be further established. Besides, the background in other NILs was complicated and they need to backcross. The identification of these NILs paves the way for novel gene cloning and provides the opportunity for gene-gene interaction analysis by constructing double or triple mutants in the same background.
