美国稻品种Lemont花粉育性中性基因Sc-n的鉴定

花粉败育是水稻籼粳杂种F1普遍存在的现象,严重制约了其杂种优势的利用。至少有Sa、Sb和Sc等6个基因座位控制籼粳杂交F1花粉育性,每个座位存在的花粉育性中性基因,可望克服其杂种F1的花粉不育性,所以发掘籼粳杂种F1花粉育性中性基因具有重要的意义。本文用美国稻品种Lemont作为待测材料,与台中65及其携带有花粉不育基因的等基因系杂交组配具有不同互作类型的杂种F1,首先观察并统计F1的花粉育性及结实率,发现Sc座位互作的杂种F1花粉育性及结实率与无互作杂种F1差异不显著;然后,采用与花粉不育基因座位紧密连锁的分子标记对该组合的F2植株基因型的分离进行检测,显示3种基因型符合1:2:1的分离比例。研究结果表明Lemont在Sc座位携带有花粉育性中性基因Sc-n,可望用于克服籼粳杂种F1的部分花粉不育,值得进一步研究。

Pollen abortion is a common phenomenon in indica and japonica hybrid F1, which extremely restrict the utilization of heterosis. There are at least six gene loci, including Sa, Sb and Sc controlling pollen fertility of hybrid F1 of indica and japonica. The pollen fertility neutral genes in the six loci maybe overcome the pollen sterility of hybrid F1. Therefore, it's very important to excavate the neutral genes in each locus. In the present study, an American rice variety, Lemont, was used for pollen fertility neutral genes detection, which was crossed with Taichung 65 and its pollen sterile near-isogenic lines. The F1s with different pollen sterility loci interactions were obtained from the test cross. Firstly, the pollen fertility rate and setting rate were investigated and was found that there is no significant difference in them between Sc interacted hybrid F1 and no pollen sterility loci (Sa、Sb and Sc) interacted hybrid F1, suggesting that there was maybe pollen fertility neutral gene in Sc locus. Then SSR markers that are closer linkage with Sc locus was used for detecting F2 population and the result showed that the segregation rate of three genotype ( Sc-i Sc-i, Sc-i Sc-j, Sc-j Sc-j ) was conformed with 1:2:1. These results demonstrated that Lemont contained a neutral gene"Sc-n"in Sc locus, which may play an important role to overcome pollen semi-sterility caused by Sc interaction of indica and japonica hybrid F1 .
