ρ因子非依赖型终止子(内源性终止子)依靠其发夹以及下游的U-tract结构促使转录过程的终止,可以有效避免下游基因转录通读,同时提高RNA聚合酶的利用效率,提高基因表达效率。然而目前研究者对终止子强度相关的序列特征的认识尚不充分,并且终止子对上游基因的影响也不明确。因此,亟待通过大范围终止子的表征,完善构效关系信息。在本研究中,我们表征了在20种具有和不具有典型的终止子结构特征序列的调控作用下,各自上下游基因的表达情况,并比较了这两种结构序列对基因表达情况影响的差异。随后,通过构建终止子突变文库,结合FlowSeq技术,收集了808条终止子序列的基因和表型特征。对其中高质量的225条终止子序列的研究表明,在不同强度的终止子的调控作用下,上下游基因的表达情况呈现反比例的趋势。最后,我们对终止子突变文库中的强弱终止子进行了划分,对其的序列特征及热力学参数分析表明终止子的茎长及U-tract区域的U碱基含量对终止子的强度具有重要的影响。本研究的结果对与理性开发终止子,并用于精准调控基因表达提供了基础信息。

he rho factor-independent terminator (intrinsic terminator) relies on its hairpin and the U-tract structure to promote the transcript termination, which can effectively avoid the read-through of downstream gene transcription and improve the utilization of RNA polymerase efficiency, thereby facilitates the regulation of gene expression.It is important to understand the key features of terminators associated with terminating efficiency, besides it is not clear if the terminator affects the upstream gene expression. Therefore, it is important to correlate the genoype and phynotype of terminator based on vast terminator date set. In this study, we first characterized the expression of the upstream and downstream gene under the regulation of 20 sequences with and without terminator structural features, and compared the effects of these two types of sequences on gene expression. Next, we constructed a terminator mutation library and collected the geno/pheno-type information of 808 terminator sequences with FlowSeq technology. 225 terminator sequences with more than 5 reads detected showed that under the regulation of terminator, the expression of upstream and downstream genes showed an inversely proportional trend. Finally, we divided the strong and weak terminators in the terminator mutation library,and the discrimination features including both sequence characteristics and thermodynamic parameterswere conducted, and the result showed the stem length and the U content of the U-tract region in the terminator have an important impact on the strength of the terminator. The informaton obtained here can be applied in the rational desgin of terminators for accruate gene expression.
