严重烫伤大鼠肝脏库普弗细胞脂多糖受体CD14膜蛋白的表达及作用

目的:研究脂多糖受体CD14膜蛋白在严重烧伤大鼠肝脏库普弗细胞中的表达情况,并初步观察对下游促炎因子TNF-α和IL-1β的表达。方法:以烫伤大鼠为模型,通过免疫组化和Western-blot 法观察严重烫伤后大鼠不同时相点肝脏普弗细胞脂多糖受体CD14膜蛋白的表达情况;ELISA检测肝细胞匀浆上清中TNF-α和IL-1β水平含量。结果:烫伤后大鼠肝脏CD14表达较假烫组相比随时间呈逐渐增强趋势,在9h达到高峰,但在12h表达开始减弱,但仍明显高于正常水平(p < 0.05);ELISA结果表明,大鼠肝组织促炎性细胞因子TNF-α和IL-1β的蛋白表达在烫伤后早期12h内,与假烫组差异明显,呈逐渐增高趋势。结论:CD14可能参与了烫伤后的炎症反应,为下一步研究了解LPS-CD14-P38MAPK 轴信号通路是否参与了烧伤后全身炎症反应中的作用机制奠定了基础。
Objective: In this paper to study lipopolysaccharide receptor CD14 membrane protein in severe burns expression in rat liver Kupffer cells, and preliminary observations the expression of the downstream proinflammatory of TNF -α and IL -1β. Method: Depend on scalded rat model, observed by immunohistochemistry and Western-blot analysis the liver Pufu cell lipopolysaccharide receptor CD14 membrane protein expression on severe burned rats in different time points, ELISA for detection of liver cell homogenate supernatant of TNF-α and IL-1β in content. Results: CD14 expression of rat liver after burn compared with the sham scald group gradually increasing trend over time, peaked at 9h, the expression began to weaken at 12h, but still significantly higher than normal levels (p <0.05). The results of ELISA show that the expression of promote inflammatory cytokines TNF - α and IL -1β have significant differences compared with fake group in scalded rat liver tissue in 12h, and to display gradually increasing trend. Conclusions: CD14 may be involved in the inflammatory response after burns, laid the foundation for further research to understand the LPS-CD14-P38MAPK shaft signaling pathway in the mechanism of the systemic inflammatory response after burn.
