基于ATP引发的滚环扩增合成DNA酶可视化检测菌落总数
Visual detection of the total bacteria by ATP triggered rolling circle amplified synthesis of DNAzymes
菌落总数是食品检疫中重要指标之一,快速、准确地对食品中的菌落总数作出判定具有重要的卫生学意义。本实验利用T4 DNA连接酶催化线性DNA模板成环需要细菌中的能量ATP参与,结合滚环扩增放大作用和DNA酶催化作用,建立了可视化检测菌落总数的新方法。优化实验条件,在最优实验条件下,菌落总数在8.2到8.2×107 CFU/mL范围与比色信号呈线性关系,检出限为6.0 CFU/mL。
he total bacteria is one of the most important indicators of food quarantine, quickly and accurately make judgement on the total bacteria has important significance in food hygiene. Utilizing ATP of bacterias as an indispensable cofactor for T4 DNA ligase to catalyze the ligation of linear DNA template, this method integrates polymerase-mediated rolling circle amplification and DNAzymes-catalyzed colorimetric reaction to visual assay for the total bacteria. Under optimal the experimental conditions, a linearrelationship between concentration of the total bacteria and colorimetric signal in the range of 8.2~8.2×107 CFU/mL with a limit of detection of 6.0 CFU/mL.
