铁皮石斛多糖抗肿瘤作用的研究

Studies on the Inhibiting Tumor Effect of Polysaccharides from Dendrobium candidum

为研究铁皮石斛多糖对人HepG2肝癌细胞,小鼠H22肝癌细胞的增殖抑制作用和对荷瘤小鼠免疫调节作用。 采用MTT法检测铁皮石斛多糖对人HepG2肝癌细胞增殖的抑制作用;小鼠接种H22实体瘤或H22腹水瘤后随机分组,灌胃不同剂量[50、100、200mg/(kg.d)]的铁皮石斛多糖,计算铁皮石斛多糖对小鼠H22实体瘤的抑制率,H22腹水瘤小鼠的存活时间以及荷瘤小鼠的内脏指数。结果表明铁皮石斛多糖对HepG2较好地抑制作用,且呈一定的剂量依赖性,多糖浓度为640ug/ml,48h,72h抑制率分别为29.3%,30.1%;铁皮石斛多糖能有效地抑制小鼠H22实体瘤生长,提高荷瘤小鼠胸腺及脾脏指数,低、中、高剂量的多糖抑制率分别为18.12%、34.78%、45.65%;中、高剂量的多糖可显著延长H22腹水瘤小鼠的生存时间。

o explore the anti-tumor activity of Dendrobium officinale polysaccharide(DOP) on HepG2 or H22 Tumor mice, and the immunoregulation effect on H22 tumor mice. Inhibition of DOP on HepG2 was determined with an MTT assay. H22 tumormice were administered orally with 50, 100, 200 mg/(kg.d) dosage of DOP, then the tumor inhibition rate, life time, organ index were measured. The result showed that HepG2 in vitro were seriously by DOP and the inhibitory was concentrations-related,640ug/ml dosage of DOP the inhibition rate was 29.3%, 30.1% when the treatment time was 48h and 72h. DOP can increase the thymus index and spieen index, and the tumor inhibition was (18.12%,34.78%,45.65%),respectively ,moreover DOP can evidently prolong life time of H22 tumor mice.
