家兔髋脱位模型中髋臼软骨病理改变研究
Pathological changes of the acetabular cartilage in the dislocation hip of the rabbit model
目的 观察家兔髋关节脱位模型中髋臼软骨的病理改变。方法 建立髋关节脱位的动物模型,通过X光片测量髋臼指数,HE染色观察组织病理改变。TUNEL法检测软骨细胞的凋亡情况。结果 固定8周后,脱位侧髋关节的髋臼指数与固定前相比明显增大(P<0.05);髋臼软骨细胞的大小及排列均不规则,软骨基质中可见多个空泡样结构;肥大层中凋亡细胞明显低于对照组(p<0.05)结论 髋关节脱位可导致髋臼软骨组织发育过程出现变化,髋臼软骨中肥大层软骨细胞的凋亡数目减少,软骨细胞堆积,可能导致软骨层增厚。
Objective To invest the pathologic changes in the acetabular cartilage of dislocated hip. Methods Using HE staining to view pathohistologic features of dysplastic acetabulum harvested from the disclocated or dysplastic hip of rabbit model which has been verified by the X-ray film. Apoptotic bodies and their distribution in the cartilage was observed by using terminal deoxynucleotidyl transferase Dutpnick-end labeling(TUNEL) staining. Results After 8 week of fixation of extended knee in rabbit, the fixation side of hip was induced dysplasia or dislocation of hip. The acetabular index was increased in dysplastic hip than contralateral hip (P<0.05). In acetabular cartilage of the control side, the apoptotic bodies widely distributed in hypertrophic zone, and some in proliferating zone, only few scattered in resting zone. In dysplastic side of the hip, acetabular cartilage contained only a few apoptotic bodies in the resting zone and superficial of proliferating zone. Conclusion The amount of apoptotic chondrocyte in acetabular cartilage of dysplastic hip was much fewer than that in the control side. The decreasing of the apoptosis in hypertrophic zone of the dysplastic acetabulum maybe causes the thickening in acetabular cartilage of the displastic hip.
