探讨自噬在小麦叶锈菌侵染结构中的形成特点及其在叶锈菌侵染小麦过程中的作用。利用自噬特异标记物MDC染色辅以激光共聚焦扫描显微镜观察,检测在叶锈菌各种侵染结构形成中是否有自噬产生;结合电子显微镜观察进一步确定自噬形成的特点。将自噬抑制剂3-MA注射到小麦叶片内,观察自噬抑制后对叶锈菌生长的影响。结果表明,将叶锈菌夏孢子接种于小麦叶片后,在叶锈菌形成的不同侵染结构中如芽管、附着胞、气孔下囊、胞间菌丝及吸器内很少观察到自噬体,只在吸器母细胞中观察到了大量自噬体;注射3-MA后大大抑制了菌丝团扩展面积及后期产孢能力。据此我们认为,吸器母细胞中的自噬对叶锈菌的生长及致病性至关重要。

In this paper,we systematically investigated the formation characteristics of autophagy in leaf rust fungus and the function of autophagy in process of leaf rust infecting the wheat. By using autophagy specific marker MDC staining and confocal observation to detect whether the leaf rust fungus infection structures have autophagy, combining with electron microscopy observation to conform the characteristics of autophagy information. The affection of autophagy to leaf rust fungus growth was observed following the injection of 3-MA(an inhibitor of autophagy synthesis). The results showed in the different structures of leaf rust fungus formation, autophagy could be observed rarely in germ tubes, appressoriums, sub-stomatal vesicles, intercellular hyphas and haustorium except haustorial mother cell. The expanding area of intercellular hyphas and the ability of spores formation were deeply inhibited by 3-MA. So we believed that the autophagy in haustorial mother cell played an important role in leaf rust fungus infection.
