苦马豆素对小鼠细胞免疫功能的影响
Effects of swainsonine on cellular immune function in mice
目的:探索SW与细胞免疫功能之间的关系,为SW的免疫毒理学研究提供新的研究思路。方法:将80只小鼠随机分为8组,5个试验组按0.05、0.2、0.8、3.2和6.4 mg/kg灌服含SW的生理盐水,连续21 d,检测淋巴细胞的增殖活性、脏器指数、血液细胞等;另3组为生理盐水、环磷酰胺和正常对照组。将正常组小鼠脾淋巴细胞,分别与0.002、0.02、0.2、2和4 μg/mL的SW共培养,检测淋巴细胞的增殖活性。结果:随着SW浓度增加,小鼠脾脏淋巴细胞未受丝裂原刺激和ConA作用时SI值先增大后减小,PHA-P作用时SI值呈剂量依赖性减小;LPS作用效果不明显;胸腺指数先增大后减小;血细胞数量减少。结论:SW剂量大于6.4 mg/kg,连续21 d灌胃,对小鼠细胞免疫功能起负面影响;SW浓度超过4 μg/mL对淋巴细胞的增殖功能起负面影响;低浓度SW对淋巴细胞功能具有一定的增强作用。基于SW不同剂量和同一作用剂量时对不同丝裂原刺激时结果不同,推测,SW 对免疫功能的影响具有选择性和双向调节作用,该作用与SW的剂量有关。
Objective:The purpose was to study the relationship of cellular immune function with different doses of SW in mice. All these can provide new thoughtful method for the study of SW on immuno-toxicology. Method:Eighty mice were divided into 8 groups randomly. Proliferative responses of spleen lymphocyte, immune organs index, cellular numbers were detected with 0.05、0.2、0.8、3.2 and 6.4 mg/kg SW by intragastric administration for 21 days. Three other groups were physiologic saline, cyclophosphamide and normal groups. Proliferative response of normal spleen lymphocyte were detected with SW doses of 0.002、0.02、0.2、2 and 4 μg/mL. Result:The stimulation index value (SI) of spleen lymphocyte with unstimulated and stimulated to ConA was increased with low doses of SW and decrease with high doses of SW. The tendency was the same as thymus index. Proliferative responses of lymphocyte stimulated to PHA-P were decreased with dose dependent of SW, but not sensitive to LPS. Cellular numbers were decreased. Conclusion:SW depressed cellular immune function by intragastric administration for 21 days at the dose over 6.4 mg/kg;SW depressed the proliferative response of spleen lymphocyte directly at the dose over 0.02 μg/mL. SW improved the function of lymphocyte to some degree at low dose of SW. These results suggest that SW can influence immuno-function by selectivity and two-way regulation with suitable doses of SW.
