利用ISSR标记分析薯蓣炭疽病菌的遗传多样性

Study on the Genetic Diversity of the strains of Anthracnose from Dioscorea by ISSR

本研究应用ISSR分子标记分析分离自薯蓣的炭疽病菌的遗传多样性。筛选出了12个ISSR引物对来自重庆、贵州、云南、福建、湖南、广西、四川等省的炭疽病菌进行PCR检测,12条ISSR引物共扩增出197条带谱,平均每个引物扩增出16.42个条带,引物的多态位点百分率达到100%。用NTSYS软件聚类分析的结果表明炭疽病菌的遗传相似性系数范围为0.61~0.94,在遗传相似系数为0.70处,可将88个菌株分为六个类群。用Genalex 6.2 软件分析,多态性位点百分比为98.42%,平均差异等位基因(Na)为1.990,有效等位基因(Ne)为1.432,Shannon指数(I)为0.415,Nei'基因多样性(h)为0.266,无偏Nei基因多样性(uh)为0.269。

In this research, analysis the genetic diversity which separated from yam application of the anthracnose of yam by ISSR molecular marker. Selected 12 primers of ISSR to PCR the strains of anthracnose from Chongqing, Guizhou, Yunnan, Fujian, Hunan, Guangxi, Sichuan Province. Received a total of 197 loci, including 196 polymorphic loci, accounting for 99.49%. Used NTSYS software to cluster, results showed that the genetic similarity coefficient of colletotrichum gloeosporioides ranged from 0.61 to 0.94, the genetic similarity coefficient was 0.70. The 88 strains could be divided into six groups. Genalex 6.2 software was used to analyze, polymorphism percentage was 98.42%, the average difference in allele (Na) 1.990, the effective number of alleles was 1.432 (Ne), Shannon index (I) was 0.415, Nei's gene diversity (H) was 0.266, unbiased Nei gene diversity (Uh) for 0.269.?????
