目的:通过免疫组化法观察复方浙贝颗粒联合阿霉素对K562/A02移植瘤细胞膜耐药相关蛋白P-gp、MRP、LRP表达影响。方法:将K562/A02细胞接种BALB/c-nu裸小鼠腋前皮下构建MDR移植瘤动物模型,给予复方浙贝颗粒灌胃联合阿霉素腹腔注射,实验结束后处死小鼠,剥离肿瘤,将瘤块组织制成切片,通过免疫组化结合Image ProPlus 6.0免疫组化彩色图象分析,观察复方浙贝颗粒联合阿霉素对K562/A02移植瘤组织细胞膜P-gp、MRP、LRP表达影响。结果:与阿霉素组相比,CZBG各剂量组移植瘤细胞膜所表达P-gp、MRP的IOD值明显降低(p<0.05),而未见明显LRP表达;结论:CZBG能协同阿霉素抑制K562/A02移植瘤组织细胞膜P-gp,MRP表达。

IM: To investigate the effects of drug-combination of Compound Zhe Bei Granule (CZBG) and doxorubicin on the expression of P-gp, MRP, LRP in K562/A02 cell line multidrug resistance tumor xenografts in mice. METHODS: Tumor xenografts model were established by injecting the multidrug resistance cell line K562/A02 in the axillary flank of BALB/c-nu-nu mice. Drug-combination of CZBG intragastric administration and doxorubicin intraperitoneal injection (i.p.) were given to the BALB/c-nu nude mice. The tumor xenografts were made into slice after the dissection, and the expression of P-gp, MRP, LRP in K562/A02 tumor xenografts in mice were investigated in immunohistochemical techniques. the integral optical density (IOD) of P-gp, MRP, LRP in K562/A02 tumor xenografts were measured by Image ProPlus 6.0. RESULTS: Compared with the single treatment of doxorubicin i.p group., the groups of the combination of the doxorubicin and CZBG with dosage classified by three types(high, middle, low) can decrease IOD of P-gp、MRP in K562/A02 tumor xenografts with statistical significance(p<0.05). There has no LRP expression in K562/A02 tumor xenografts in five groups. CONCLUSION:The combination of CZBG and doxorubicin can decrease the expression of P-gp, MRP in K562/A02 multidrug resistance tumor xenografts in mice.
