为研究海参皂苷酸降解的规律,制备稀有次级苷和苷元化合物,本研究选用一种典型的海参皂苷-echinoside A,采用三氟乙酸(TFA)和稀硫酸分别对其进行酸水解,采用柱层析、高效液相色谱等制备技术,获得苷元化合物3个,次级苷8个。化合物以NMR、ESIMS等手段进行结构鉴定,探讨了TFA和稀硫酸对海参皂苷降解的规律。发现TFA水解产物单一,仅生成一个二醇苷元,但苷元上会发生脱水和双键转移;稀硫酸水解echinoside A生成1-4糖一系列次级苷化合物及三醇和二醇苷元,所有化合物皆为脱硫酸酯基产物,且部分化合物发生苷元上的脱水反应,但是双键未发生转移。研究的结果将为定向制备海参次级苷和苷元化合物以及研究海参皂苷水解、消化、代谢、构效关系等方面的工作提供一定的支持和依据。

In this research we selected a classical triterpene glycoside from sea cucumber - echinoside A to investigate the hydrolysis characters of holothone triterpene glycoside by acid. Also, we investigated the preparation of rare secondary glycosides and aglycones in this paper. 2 M of trifluoroacetic acid (TFA) and 0.2 M of sulfuric acid were employed as the hydrolysis solvent. The hydrolysis products were isolated and purified by column chromatograpy, high performance liquid chromatograpy (HPLC), etc. Three aglycones and eight secondary glycosides were obtained. Their chemical structures were determined by NMR and ESIMS. Depending on their structures the hydrolysis characters were illuminated. Hydrolysis products by TFA only contained an aglycone with two -OH groups. The structure of aglycone indicated that dehydration was occurred on aglycone, and the positions of double bonds were changed. Hydrolysis products by 0.2 M of sulfated acid contained a series of secondary glycosides with 1-4 monosaccharide and aglycones with two or three -OH groups. All products did not have sulfate ester which presented in the sugar chain of original echinoside A. Half products were dehydrated on aglycone part. However, the positions of double bonds were not changed. The result will supply methods to prepare some certain secondary glycosides and aglycones. It will also support future studies on hydrolysis, digestion, metabolism and bioactivities of triterpene glycosides from sea cucumber.
