α-葡萄糖苷酶抑制剂是目前治疗Ⅱ型糖尿病的常用药物,在调节机体代谢过程中起着关键的作用。多项研究证实红茶可以有效预防糖尿病,降低餐后血糖含量,本实验系统研究了三种茶叶提取物---茶黄素(theaflavins)、茶红素(thearubigins)和茶褐素(theabrownins)对α-葡萄糖苷酶活性的影响,同时考察了它们对α-葡萄糖苷酶抑制动力学。研究结果表明, 三种茶色素均可强烈抑制α-葡萄糖苷酶活性,半抑制率分别为8.19μg/ml、4.50μg/ml和6.98μg/ml,远远低于目前治疗Ⅱ型糖尿病的常用药物阿卡波糖(IC50=1.04mg/ml),茶红素对α-葡萄糖苷酶属于非竞争性抑制,而茶黄素和茶褐素对α-葡萄糖苷酶属于混合型竞争性抑制。

α-Glucosidase inhibitors is one of the commonly used drugs for the treatment of type Ⅱ diabetes, which play a key role in regulating the body′s metabolic process. Several studies have confirmed that black tea could prevent diabetes, and reduce postprandial blood glucose levels. This experiment systematically studied the effect of three kinds of tea pigments - theaflavins, thearubigins and theabrownins on α-glucosidase activity, as well as inhibition type on α-glucosidase. The results indicate that theaflavins、thearubigins and theabrownins can strongly inhibit α-glucosidase activity, with half inhibition rate of 8.19μg/ml, 4.50μg/ml and 6.98μg/ml, which are much better than the current commonly used therapeutic agent - acarbose (IC50=1.04mg/ml)in the treatment of type Ⅱ diabetes. The results showed that thearubigins had a non-competitive inhibitory effect on α-glucosidase whereas theaflavins and theabrownins had a mixed inhibitory effect on α-glucosidase.
