平菇高产漆酶菌株的筛选

he Screening on the High Production of Laccase from 14 Pleurotus ostreatus strains

采用PDA-Bavendamm氏反应显色和氧化带显色法检测14株平菇菌株产漆酶特性,结果表明,14株平菇均具漆酶活性。通过初筛,淘汰掉酶活力较低的3株,对其中的11株平菇进行液体培养发酵,由发酵液制备粗酶液,以愈创木酚为底物,检测漆酶活性,11个菌株大多在12d左右达到产酶高峰,6个菌株表现了较高的漆酶活性,其中,曲师911和桂黑5号漆酶最大酶活分别达到70.03U/ml和51.96U/ml。

olorful reactions of 15 Pleurotus ostreatus strains were studied through PDA- Bavendamm plate and PDA- guaiacol plate separately.It was proved that 14 Pleurotus ostreatus strains could all produce laccase.By screening out 3 strains with low enzyme activity of laccase,the rest 11 Pleurotus ostreatus strains were cultured in liquid medium.Crude enzyme were made from the fermentation broth,and the enzyme activity of laccase by using guaiacol as the substrate.Most of 11 strains took 12 days to reach the peak of enzyme production,among them, the enzyme activity of Qushi 911 and Guihei No.5 arrived 70.03U/mL and 51.96U/mL respectively.
