目的:基于抗体蛋白芯片技术筛选慢性应激抑郁模型大鼠海马组织差异蛋白,观察电针对血管内皮生长因子-C(VEGF-C)、趋化因子受体-4(CXCR4)和趋化因子IP-10表达的影响,进一步探究电针抗抑郁机制。方法:采用慢性应激抑郁大鼠模型,将40只SD大鼠随机分为正常组、模型组、电针组和氟西汀组。电针组于每日给予应激前 1 h电针"百会"和"印堂"穴;氟西汀组于每日给予应激前1 h灌胃。应用抗体蛋白芯片技术检测海马VEGF-C、CXCR4和趋化因子IP-10蛋白表达的变化,结果以fold change来判断蛋白的差异表达。结果:与正常组比较,慢性不可预知性应激可使模型组大鼠海马VEGF-C、CXCR4和趋化因子IP-10表达明显上调(fold change = 1.34;1.21;1.50);与模型组比较,电针和氟西汀逆转了海马VEGF-C、CXCR4和趋化因子IP-10的上调表达(fold change = 0.66,0.65,0.72;fold change = 0.59,0.57,0.62)。结论:电针对慢性不可预知性应激抑郁状态的治疗作用可能是通过调节海马VEGF-C、CXCR4和趋化因子IP-10的表达来实现的。

Objective: To investigate the effect of electro-acupuncture (EA) on hippocampal vascular endothelial growth factor C (VEGF-C), chemokine cys-x-cys receptor 4 (CXCR4) and interferon-inducible protein-10 (IP-10) in depression disorder rats. Methods: This rat model of depression disorder was induced by unpredictably chronic-mild stimulation. Forty male rats were randomly divided into normal, model, EA and fluoxetine groups. Rats in EA group were elecro-acupunctured at GV20 (Baihui) and GV29 (Yintang). Rats in fluoxetine group were administered with fluoxetine (0.18 ㎎/ml) by gavage (1ml/100g). At 28 days post-intervention, the expression of hippocampal VEGF-C, CXCR4 and IP-10 were detected based on antibody microarrays technology. Fold change, ratio of the density of the proteins, was used to investigate alteration of the differential protein expression. Results: Compared with the normal group, unpredictably chronic-mild stimulation up-regulated the expression of hippocampal VEGF-C, CXCR4 and IP-10 in model group (fold change = 1.34; 1.21; 1.50). Compared with the model group, EA and fluoxetine reversed the up-regulation of hippocampal VEGF-C, CXCR4 and IP-10 (fold change = 0.66, 0.65, 0.72; fold change = 0.59, 0.57, 0.62). Conclusion: There is a possibility that the effects of electro-acupuncture on depression disorder is to regulate the expression of hippocampal VEGF-C, CXCR4 and IP-10, which might elucidate the mechanism of the anti-depressive effect of EA therapy.
