水稻耐盐萌发能力的遗传分析和QTL定位
Genetic and QTL analysis for germination ability under salt stress condition in rice (Oryza sativa L.)
利用韭菜青/IR26 F2:9重组自交系(RILs)群体构建分子遗传图谱,并在100 mmol/L的NaCl胁迫下处理10 d鉴定种子的萌发能力。采用主基因-多基因混合遗传模型和复合区间定位法,对水稻耐盐萌发能力进行遗传分析。结果表明,水稻耐盐萌发能力的次数分布呈连续变异,是由多基因控制的数量遗传性状。由2-3对主基因+多基因控制水稻耐盐萌发性状,以主基因遗传为主,发芽率、发芽指数、芽长和种子活力主基因遗传率高,均超过60%。在第2、4、6、9染色体上共检测到5个QTLs:吸胀速率2个(qIR-4、qIR-9)、发芽率2个(qGR-2-1、qGR-2-2)、种子活力1个(qSV-6)。QTL表达存在时效性,单个QTL能解释9.6 - 46.6%的表型变异,其中主效QTL qGR-2-1能解释46.6%的表型变异。本研究检测到的大部分QTLs与已报道的水稻耐盐相关QTLs位置相同或相近,表明结果真实可靠;未发现与qIR-4位置相近的QTL,可能为新检测的耐盐基因座。
One F2:9 recombinant inbred lines (RILs) population, derived from a cross between IR26 (Oryza sativa L. spp. indica) and Jiucaiqing (japonica), was used to construct a molecular linkage map and to identify the germination ability under 100 mmol/L NaCl for 10 d in this study. A major gene plus polygene mix inheritance model and a composite interval mapping approach were applied to conduct genetic analysis for germination ability. The frequency distributions of germination traits under salt stress in RILs population showed continuous segregation, suggesting these were quantitative traits controlled by several genes. The germination traits were complex regulated by two or three major genes plus polygene and the traits were mainly dominated by major genes, among the traits of germination rate, germination index, shoot length and seed vigor with high heritability value (over 60%). Five QTLs were identified on chromosome 2, 4, 6 and 9: two for imbibition rate (qIR-4 qIR-9), two for germination rate (qGR-2-1 qGR-2-2) and one for seed vigor (qSV-6) respectively. The phenotypic variance explained by a single QTL ranged 9.6 - 46.6% and one major QTL qGR-2-1 accounted for 46.6% of the phenotypic variation. The expression of QTLs were developmentally regulated and growth stage-specific. Most of QTLs observed here shared the similar regions of QTLs for rice salt tolerance reported previously, indicating these QTLs were reliable. However, qIR-4 was firstly reported here.
