Preliminary study on Hela cell treated by Pinellia ternata lectin

本研究利用甘露糖-Sepharose 4B亲和柱从半夏总蛋白中分离纯化得到一种12 KD的半夏凝集素,经研究表明其为半夏凝集素单体。将该凝集素作用于体外培养的HeLa细胞,MTT法检测细胞活性并用倒置相差显微镜观察凝集素对细胞生长状态的影响。结果显示:利用甘露糖-Sepharose 4B亲和柱能得到较高纯度的半夏凝集素单体,低浓度(0.004 mg/mL、0.02 mg/mL、0.1 mg/mL)半夏凝集素具有促进HeLa细胞增殖的作用,且随浓度增高促增殖作用减弱,随时间增加促增殖作用减弱;高浓度(0.5 mg/mL、1 mg/mL)半夏凝集素有抑制Hela细胞增殖的作用。实验结果反映出半夏凝集素对HeLa作用有时间和剂量依赖性。

In this paper, one kind of Pinellia ternata lectin was isolated from total protein of Pinellia ternata by using mannose-Sepharose 4B affinity chromatography. Cultured Hela cells adding the lectin by different concentrations was detected by MTT method and observed with inverted phase contrast microscope. The results showed that: high purity lectin was received, low concentrations (0.004 mg/mL, 0.02 mg/mL, 0.1 mg/mL) Pinellia ternata lectin could promote Hela cell proliferation, and the effect weakened with the concentration and time increased; high concentrations (0.5 mg/mL, 1 mg/mL) Pinellia ternata lectin inhibited Hela cell proliferation, and this effect increased in a time and dose-dependent manner.
