目的:初步观察"证侯"对附子"毒性"的影响,为临床辨证用药提供实验依据。方法:以冰水游泳法复制寒证模型。小鼠冰水游泳后,灌胃给予生附片水煎液,用Bliss法测定LD50;大鼠冰水游泳后静脉注射乌头碱,描记体表心电图,计算出现心律失常的时间。结果:冰水游泳后实验动物出现呼吸微弱,寒战,畏寒喜暖,蜷缩少动,舌、耳、爪、尾部黯淡等寒证表现。Bliss法求得游泳小鼠的LD50、LD95较未游泳小鼠有所升高,而冰水游泳大鼠出现心律失常的总时间较未游泳大鼠明显缩短。结论:冰水游泳寒冷刺激在某种程度上可模拟中医寒证的某些表征,附子及乌头碱对寒证动物的毒性有所降低,"证"和"毒"具有一定的相关性。

Objective: To observe the effect of syndrome on the toxicity of aconite roots, in order to provide experimental basis for treatment based on syndrome differentiation. Methods: To establish animal model of cold syndrome by ice-water swimming. After swimming, water decoction of crude aconite roots was administrated to mice by gavage, and then LD50 was determined by Bliss method. After swimming, aconitine was administrated to rats by vein injection, and then calculated the duration of arrhythmia. Results: After ice-water swimming, mice and rats appeared cold syndrome, such as weak breathing, shivering, intolerance of cold, preference for swarming, quiet, dull tongue, dull ears, and so on. Compared with the non-swimming mice, LD50 and LD95 of the swimming ones increased slightly. Compared with the non-swimming rats, the duration of arrhythmia of the swimming ones was shorter. Conclusion: Ice-water swimming could simulate cold syndrome to some extent. The toxicity of aconite roots and aconitine to the animals of cold syndrome decreased, which suggested that syndrome and toxicity has correlation.
