五个菌草新品种rDNA ITS序列克隆与遗传多样性分析
Phylogenetic analysis of 5 new cultivated JUNCAO varieties
对从野生环境中筛选出五个优质的菌草品种的5.8S rDNA、ITS1及ITS2片段进行克隆及序列分析,并登入Genbank数据库进行比对,采用DNAMAN、MEGA等软件分析其遗传关系的聚类图。结果显示,克隆的目的片段长度为634~642 bp之间,聚类结果反映出其与相识品种之间遗传距离,并结合形态学鉴定方法初步确定JC01、JC02、JC03、JC04为芦竹属,JC05为狼尾草属,且与库中现有品种存在较大差异,可能为新品种。
Phylogenetic relationships among 5 cultivated JUNCAO varieties screened from the wild was studied by ITS1-5.8s-ITS2 rDNA sequences. Results showed that the length of cloning sequences are 634-642bp, and the genetic relationship amount 5 JUNCAO strains were analyzed by cluster. Combined with the results of the morphological identification, We preliminarily confirm that JC01,JC02,JC03 and JC04 is Arundo and JC05 is Pennisetum. They are quite different from existing varieties and therefore may be new cultivated JUNCAO varieties.
