多肽类药物的口服吸收是世界性难题,为了探索和研究如何利用蚕蛹粉中某些特定组分对提高目的蛋白的口服吸收利用度,本课题建立了蚕蛹粉中30 kDa蛋白群以及壳聚糖的制备工艺标准,利用蚕蛹粉30 kDa蛋白群与壳聚糖所构成的轭合物包裹重组人粒细胞-巨噬细胞集落刺激因子(rhGM-CSF)灌胃促进小鼠对rhGM-CSF的吸收以及提高增强型绿色荧光蛋白(EGFP)被轭合物包裹后在人工消化液中的稳定性。实验结果表明,30 kDa蛋白群与壳聚糖的轭合物通过包裹rhGM-CSF再给小鼠灌胃,ELISA检测血清中的rhGM-CSF含量比单独将rhGM-CSF灌胃给小鼠所测量得到的吸收值高15倍以上;利用荧光显微镜观测与轭合物充分混合的EGFP在37℃条件下的人工消化液中的稳定性要比未结合轭合物的EGFP的稳定性高。因此证明家蚕蛹中30 kDa蛋白群与壳聚糖的轭合物确实能提高蛋白质的口服吸收利用度,并且有保护目的蛋白避免消化液降解的作用。

he oral absorption of peptide drugs is a worldwide problem, in order to increase the degree of oral absorption, we studied in and separated the silkworm pupa powder to find out some special components, our research established the preparation standard of the 30 kDa proteins and chitosan which were made from the silkworm pupa powder, using the conjugates jointed by the 30 kDa proteins and chitosan to wrap the recombiant rhGM-CSF, and fed the mice with the mixture in order to increase the degree of oral absorption, we also let the conjugates wrap with EGFP been digested in the artificial digestion, detected the stability. The result shows by the ELISA detection we found that 15 times higher the amount of rhGM-CSF in the experiment group than in the negative control group; With the fluorescence microscope we found that the EGFP mixed with the conjugates and digested in the artificial digestion(at 37℃) had more stabilize than the EGFP without the conjugates. The summary: our research proved the conjugates(made up with the 30 kDa proteins and chitosan) can increase the degree of oral absorption, and also the proteins which been mixed with the conjugates would avoid from the digestion juice.
