左氏无绿藻对奶牛乳腺上皮细胞先天性免疫应答的影响

左氏无绿藻是一种导致奶牛乳腺严重感染的乳房炎病原菌。然而,奶牛对左氏无绿藻感染的先天免疫应答机制尚不清楚。因此,本文旨在研究体外左氏无绿藻感染的先天免疫应答。通过对奶牛乳腺上皮细胞感染5.0×104 个/ml菌液的基因II型左氏无绿藻,于1,3,6,12,24小时后通过荧光定量PCR检测TLR-2,TLR-4,TNF-α,IL-1β,IL-8,NOD-1,NOD-2和β-defensin-5的mRNA表达水平变化,同时检测相应时间被感染奶牛乳腺上皮细胞的细胞质和细胞核中NF-κB p65蛋白表达水平。结果显示基因II型左氏无绿藻侵染细胞后,TLR-4,TNF-α,IL-1β,IL-8,NOD-1,NOD-2和 β-defensin-5的mRNA表达水平与对照组相比在一定时间点内上调。Western Blot试验结果显示当乳腺上皮细胞受到基因II型左氏无绿藻感染时,NF-κB p65蛋白的表达在细胞核内增加,在细胞质中减少。结论:乳腺上皮细胞感染II型左氏无绿藻可在一定程度上调节先天性免疫应答和NF-κB介导的炎症信号通路,从而导致奶牛发生乳房炎。
Prototheca zopfii is an important bovine mastitis pathogen, which could result in severe mammary infection. However, the innate immune response in bovine mastitis associated with P. zopfii was not clear. Therefore, the aim of this study is to investigate invitro innate immune responses implicated by P. zopfii.Bovine mammary epithelial cells (bMECs) were infected with5.0 x104 cells/ml P. zopfii genotype-II independently and the mRNA expression of TLR-2, TLR-4, TNF-α, IL-1β, IL-8, NOD-1, NOD-2 and β-defensin-5 were determined by real-time polymerase chain reaction (RT-PCR) over a time course of 1, 3, 6, 12 and 24h. The detection of the NF-κB p65 protein in nucleus and cytoplasm of infected bMEC over the same time course were evaluated. Results showed that P.zopfii genotypes-II hasability to up-regulate the expression of TLR-2, TLR-4, TNF-α, IL-1β, IL-8, NOD-1, NOD-2 and β-defensin-5 more strongly than the control group. Western blotresults showed that when bMECs were challenged byP.zopfii genotype-II, the expression of NF-κB p65 protein in the nucleus was up-regulated while in cytoplasm it appeared to be repressed, which indicated that bMECs partlyregulate the innate immune responses and inflammationby the NF-κB signaling pathway whilst being infected by P. zopfii genotype-II. It was concluded that genotype-II causes inflammation of bovine mammary tissue.
