青鲜素对大鼠肝脏氧化-抗氧化系统平衡的影响

目的 探究青鲜素对大鼠肝脏氧化应激的影响,从氧化损伤的角度探讨其对机体的毒作用机制。方法 将40只清洁级SD大鼠,随机分为4组,分别为空白对照组和低剂量(125mg/kg)、中剂量(250 mg/kg)、高剂量(500 mg/kg)青鲜素染毒组,每组10只,雌雄各半。采用经口灌胃方式进行染毒,每天1次,连续染毒14天。末次染毒24h后测量大鼠体重和脏器湿重,并计算脏器系数;测定大鼠肝组织匀浆中超氧化物歧化酶(SOD)、谷胱苷肽过氧化物酶(GSH-Px)活性及丙二醛(MDA)含量。结果 与对照组相比,500mg/kg青鲜素染毒组,SOD活性降低了78.7%,且差异具有统计学意义(P<0.05);250 mg/kg、500mg/kg青鲜素染毒组,GSH-Px活性分别降低47.4%、48.3%,且差异有统计学意义(P<0.05);125 mg/kg、青鲜素染毒组MDA含量上升,但差异无统计学意义(P>0.05)。结论 青鲜素染毒对大鼠肝脏系统存在毒性作用,可能是通过影响肝脏组织中SOD和GSH-PX的活力和破坏氧化-抗氧化系统的平衡等导致肝脏的损伤。

objective: To study the effects of Maleic Hydrozine on oxidative stress in liver of rat, to explore its mechanisms of toxicity to the rat in the perspective of oxidative damage. Methods: 40 healthy SD rats were randomly divided into four groups: blank control group, low-dose group, moderate-dose group, high-dose group. The rats were exposed to MH through mouth, once a day for two consecutive weeks. The four groups were treated with dosages of 125mg/kg, 250mg/kg, 500mg/kg. 24 hours after the 2 weeks treatment, measure the weight and wet weight of the organs, calculate the ratio of the organs. Measure the content of malondialdehyde(MDA), the activity of superoxide dismutase(SOD), glutathione peroxidase(GSH-Px) in liver. Results: The activity of SOD and GSH-Px was significantly lower than those in the control group(P<0.05).The activity of SOD decreased by 78.7% at 500mg/kg dose levels; the activity of GSH-Px decreased by 47.4%、48.3% at 250mg/kg,500mg/kg dose levels; the content of MDA increased, but not significantly(P>0.05).Conclusion: MH exposure damages the rat's liver, the mechanisms of toxicity may be that MH exposure breaks the balance of the oxidant-antioxidant system and effects the activity of the SOD and GSH-Px in the liver.
