为了利用原生质体技术选育茂原链霉菌(Streptomyces mobaraensis)11019的高产谷氨酰胺转氨酶菌株,摸索该菌株原生质体制备和再生的条件。结果表明,茂原链霉菌11019原生质体制备和再生的最佳条件为:在培养菌丝体的培养基中添加0.05%的甘氨酸,培养温度为28℃,培养时间为28h,溶菌酶浓度为2.5mg/ml,酶解温度为33℃,酶解时间为1.5h.-80℃低温保存时,有效保护剂为:5%DMSO+2%蔗糖+0.5mol/l甘露醇。

In order to screen a high-yielding transglutaminase strain from Streptomyces mobaraense 11019,protiplasts preparation and regeneration of the strain were optimized.The results showed that the optimum conditions fpr the preparation and regeneration of the protoplasts were as follows:concentration of glycine were at 0.05%,culture temperature 28℃,culture for 28 h:lysozyme 3mg/ml,enzymolysis temperature 33℃,and enzymolysis for 1.5h.The optimum protective agent is 5% DMSO+2% sucrose+0.5 mol/l mannitol in low temperature storage.
