miRNA在植物正常生长发育过程中发挥重要的调控作用。有众多研究报道,植物病毒的侵染会导致植物内源miRNA表达谱的变化,这些变化被认为是受侵染植物症状发生的原因之一。可是对于miRNA的表达变化对于病毒侵染有何作用,鲜有研究报道。本文分析了实验室前期鉴定的一个水稻新miRNA--nmiR119在水稻条纹病毒侵染过程中的作用。结果表明,RSV侵染会导致nmiR119的表达上调,而经验证后的两个靶基因Os10g28820和Os10g326400表达下调;获得的超表达nmiR119水稻与野生型水稻没有明显表型差异。而nmiR119功能抑制的水稻植株结实率显著下降;接毒分析发现,nmiR119超表达植株和功能抑制植株的RSV发病率相比对照均有所升高,但RSV积累量在功能抑制的植株中极显著减少,暗示nmiR119可能在调控RSV复制、增殖环节执行某种功能。

ImiRNA plays key regulation roles in the plant development. many reports have been reported that viral infection could lead to the change of plant miRNA expression pattern, which was supposed to be a reason of viral symptom development. Meanwhile, there were few reports on what the roles of miRNA expression pattern change are to viral infection. Here, a novel miRNA, nmiR119, identified previous from small RNA libraries of rice stripe virus (RSV)-infected rice by us was focused to understand its function. Results showed that nmiR119 was upregulated in the RSV-infected rice plant, and their two target genes were downregulated correspondingly. Transgenic rice plant overexpressing nmiR119 had the similar phenotype to wild type plant, while the nmiR119-suppressed rice plant showed the extreme low seed rate. In RSV challenge analysis, both nmiR119-overexpressed plant and nmiR119-suppressed plant had the slight higher incidence of disease comparing to the wild type plant. But, the accumulation of RSV RNAs were at a extreme low level, indicating the potential roles of nmiRNA played for viral RNA replication.
