苦瓜提取物抑制3T3-L1细胞脂肪沉积的初步研究
Effects of bitter melon extracts on adipogenesis of 3T3-L1 adipocytes
肥胖是一种严重危害人类健康的慢性炎性疾病,表现为脂肪过度沉积与低度炎症反应。已有研究表明苦瓜具有一定减肥效果,但其确切分子机理尚未阐明。本研究以3T3-L1 细胞3T3-L1细胞为模型,利用RT-PCR技术研究苦瓜提取物对脂肪沉积转录因子和脂肪酸代谢关键基因的调控作用。结果显示,苦瓜果实正丁醇提取物(三萜类混合物)能明显的抑制3T3-L1细胞脂肪沉积,抑制转录因子C/EBPα、PPARγ和SREBP-1c的基因表达,对脂肪酸代谢基因GLUT4、ACC1、Fasn、aP2和LPL抑制明显。本研究为开发苦瓜减肥功能性食品提供理论依据。
Obesity is recognized as a low-grade chronic inflammatory disease characterized by lipid accumulation(adipogenesis), causing serious public health problems. Bitter melon (Momordica charantia) has been shown to ameliorate diet-induced obesity, but the molecular mechanisms are still unknown.The objective of this study was to elucidate the effect of bitter melon extracts on adipogenesis in 3T3-L1 adipocytes.The results revealed that bitter melon markedly inhibited lipid accumulation. The n-butanol fraction blocked the expression of adipogenic transcription factors, including peroxisome proliferator-activated receptors (PPARγ), CCAAT/enhancer binding proteins (C/EBPα) and Sterol regulatory element binding protein-1c (SREBP-1c). Morover, down-regulated the mRNA levels of 5 key lipogenic enzyme gene expression, including FAS、GLUT4、ACC1、Fasn、aP2 and LPL. These results indicate that n-butanol fraction of bitter melon suppresses adipocyte differentiation and lipid accumulation through multiple mechanisms and may have applications for the treatment of obesity.
